摘 要:目的:评价复方鳖甲软肝方对自发性高血压大鼠(SHR)心肌纤维化、左室重构及血浆中血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)、醛固酮的效应。方法:实验选用12周龄SHR50只,随机将其分为5组,即空白对照组、依那普利组、小、中、小剂量复方鳖甲组(小、中、大剂量药物组),每组各10只。另取正常SD大鼠10只作为正常对照组。测定治疗10周后各组大鼠收缩压、心脏质量指数(heartweightindex,HWI)、左室质量指数(leftventricularmassindex,LVMI)、胶原蛋白含量,血浆中血管紧张素Ⅱ、醛固酮含量,心肌胶原容积分数(collagenvolumefraction,CVF)和血管周围胶原面积(perivascularcircuferentialarea,PVCA)。结果:治疗10周后,空白对照组大鼠收缩压,HWI,LVMI,胶原蛋白含量犤(195±9)mmHg,(5.38±0.25),(3.81±0.09),(6.13±0.93)mg/g,1mmHg=0.133kPa犦均明显高于正常对照组犤(127±10)mmHg,(3.88±0.28),(2.57±0.17),(4.19±0.72)mg/g犦(P<0.01)。依那普利组大鼠收缩压明显低于与空白对照组(P<0.01),而复方鳖甲软肝方各组收缩压比较,差异无显著性意义(P>0.05);依那普利组大鼠HWI,LVMI明显低于空白对照组(P<0.01),而复方鳖甲软肝方各组差异无显著性意义(P>0.05);依那普利组,复方鳖甲软肝方中。AIM:To investigate the effects of biejia ruangan recipe on myocardial fibrosis,left ventricular remolding,angiotensin Ⅱ(AngⅡ) and aldosterone(ALD) in plasma in the spontaneously hypertensive rats(SHRs). METHODS:Fifty 12-week-old SHRs were randomly divided into 5 groups:SHR-C(untreated),SHR-Y(treated with enalapril),SHR-B1,B2 and B3(treated with the low,moderate and high dose of biejia ruangan recipe respectively),each group consisting of 10 rats.And 10 normal SD rats were used as negative controls.Systolic blood pressure,heart weight index(HWI),left ventricular mass index(LVMI),left ventricular collagen contents,Ang Ⅱand aldosterone in plasma,collagen volume fraction(CVF),perivascular circuferential area(PVCA) were determined 10 weeks after treatment. RESULTS:At 10 weeks after treatment,systolic blood pressure,HWI,LVMI and left ventricular collagen contents of SHR-C group[(195±9) mm Hg,(5.38±0.25),(3.81±0.09),(6.13±0.93) mg/g,1 mm Hg =0.133 kPa] were significantly higher than those of negative control group[(127±10) mm Hg,(3.88±0.28),(2.57±0.17),(4.19±0.72) mg/g](P< 0.01).The systolic blood pressure was significantly lower in SHR-Y group than in SHR-C group(P< 0.01),but was not significantly different among SHR-B1,B2 and B3 groups(P >0.05).The HWI and LVMI were significantly lower in SHR-Y group than in SHR-C group(P< 0.01),but were not significantly different among SHR-B1,B2 and B3 groups(P >0.05).The left ventricular collagen contents in SHR-Y,SHR-B2 and SHR-B3 groups were significantly lower than those in SHR-C group(P< 0.05-0.01).The contents of AngⅡand aldosterone,CVF and PVCA in SHR-C group were significantly higher than those in negative control group(P< 0.01).The content of AngⅡin SHR-Y,B2 and B3 groups was significantly lower than that in SHR-C group(P< 0.01).The content of aldosterone in SHR-Y group was significantly lower than that in SHR-C group(P< 0.01),decreased in SHR-B1 group with significant difference(P< 0.05),significantly decreased in SHR-B2 and SHR-B3 groups(P< 0.01),and s
