摘 要:目的 探讨中药溶栓颗粒对脑缺血再灌注后累积性损伤的脑保护作用 ,为缺血再灌注损伤的防治提供新的药物。方法 实验分空白对照组、模型组、阳性药物对照组、中药 1组、中药 2组 ,采用改良过的Pulsinelli的 4VO法[1,2 ] ,以放射免疫分析法测血清及脑内肿瘤坏死因子 -α(TNF -α)含量 ,同时对脑组织含水量进行测定。结果 模型组大鼠血清和脑匀浆中TNF -α含量在脑缺血再灌注后均明显高于空白组 ,中药 1组、2组预防给药后血清、脑内TNF -α含量较模型组仅有小幅度的增高。中药 1组、2组与阳性药物对照组比较有统计学意义 (P <0 .0 5~P <0 .0 1)。且随剂量增加TNF -α增高幅度越小。脑缺血再灌注模型组脑含水量与空白对照组比较有统计学意义 (P <0 .0 1) ,而中药不同剂量组及尼莫地平治疗组脑含水量与模型组比较均有统计学意义 (P <0 .0 1) ,且随剂量增加脑含水量减少。结论 溶栓颗粒对脑缺血再灌注具有保护作用 ,其机制与该方降低TNF -α的过量释放、减少脑含水量有关。Objective To investigate the protective effect of the herbal medicine Rongshuan granule (RSG) on brain with cumulative injury after ischemic reperfusion injury,and to offer new medicine for the prevention and cure of ischemic reperfusion injury.Methods In the experiment ,rats were randomly divided into five groups: normal control group、model group、positive medicine control group、herbal medicine groupI and herbal medicine groupII.With improved Pulsinelli 4VO measure,the content of tumor necrosis factor-α (TNF-α) in serum and brain,and the moisture content of brain tissuie were mensurated by radioimmunoassay analysis. Results As compared with the model group,the content of TNF-α in homogenate of serum and brain had a significantly low percentage in normal control group after cerebral ischemic reperfusion injury,and only had a little higher percentage in herbal medicine groupI and herbal medicine groupII;that both herbal medicine groupI and herbal medicine groupII both had significant or greatly significant difference compared with positive medicine control group(P<0.01),the raising level of TNF-α turned reduced with the dosage increased.The moisture content of brain tissuie undergoing ischemic reperfusion injury in model group had extremely significant difference compared with the other four groups(P<0.01),and also turned decreased with the dosage increasing.Conclusion RSG could protect the brain undergoing ischemic reperfusion injury,the function mechanism was related in the decrease of over-release of TNF-α and cerebral moisture content.
