摘 要:依次采用石油醚、氯仿、乙酸乙酯、丙酮、乙醇分级提取芫花根,以GC-MS分析各分级提取物(E1~E6)的化学成分,以小鼠网状内皮系统(RES)对刚果红的吞噬活性和小鼠毛细血管通透性为模型,评价其对急性炎症的抑制活性。E1~E6共检测出93种化合物,其中脂肪族化合物75种,芳香族化合物13种(主要为邻苯二甲酸酯衍生物),生物碱类化合物5种。其中,E中芳香族化合物的含量最高.达24.943%。E1~E6均能提升小鼠RES对刚果红的吞噬活性和显著抑制小鼠毛细血管通透性的增加,其中E的活性最为显著。邻苯二甲酸酯衍生物的存在可能是芫花根分级提取物具有抗炎活性的物质基础。E1~E6对急性炎症的抑制作用可能是通过提升RES的吞噬活性和竞争性地与组胺受体结合来实现的。The radix Daphne genkwa was sequentially extracted by petrol ether, chloroform, acetyl acetate, acetone and ethanol and the chemical constituents of the extracts (E 1 ~E 5 ) were analyzed by GC MS. The inhibition on acute inflammation of the extracts was evaluated by the enhancement to phagocytic activity of reticular epithelial system (RES) to Congo red and the inhibition on vascular permeability in mice. 93 compounds including 75 aliphatics, 13 aromatics (predominately comprised by acyl phthalate derivatives) and 5 alkaloids were identified in E 1 ~E 5 , among which the total content of aromatic compounds in E 5 was observed to be 24.943%, much higher than those in E 1 ~E 4 . On inhibition of acute inflammation, the 5 fractions presented obvious enhancement on phagocytic activity of RES and inhibition of vascular permeability. E 5 , with the highest content of aromatic constituents, indicated stronger action for inhibiting acute inflammation. The existence of acyl phthalate derivatives might be the reason for radix Daphne genkwa to possess inhibition on acute inflammation. This inhibition is likely to be realized by enhancing phagocytic activity of RES and competitively binding with histamine receptor.
