液氮冷冻对成骨细胞免疫原性影响的实验研究

摘　　要：目的 :探讨液氮冷冻对成骨细胞免疫原性的影响。方法 :体外培养成骨细胞 ,传代鉴定后液氮冻存 3个月 ,复苏。利用流式细胞仪 (FCM ) ,通过间接免疫荧光技术测定冻存前后成骨细胞的免疫原性。结果 :体外培养细胞经鉴定为成骨细胞 ,液氮冻存 3个月后存活率 >90 %。FCM测定结果表明冻存后成骨细胞的免疫原性显著降低 (P <0 .0 1)。结论Objective:To investigate the effects of f luid nitrogen cryopreserva tion on immunogenicity of osteoblasts.Method:The osteoblasts were cultured a nd examined by ALP (alkaline phosphatase) stains and calvaria stains, and then cryopreservated for 3 months. Having been labelled by immunofluorescene techniq ue,the MHC-Ⅰ antigen of osteoblasts was examined by Flow Cytometry assy. Result:Cells cultured in vitro were identified as osteoblasts. The immunogen icity of osteoblasts was greately reduced by fluid nitrogen cryopreservation( P<0.01).Conclusion:Fluid nitrogen cryopreservation is an ide al method to reduce the immunogenicity of osteoblasts. [
