摘 要:目的 :研究苯那普利联合科素亚对自发性高血压大鼠肾脏损害的保护作用。方法 :用 13月龄SHRs作为实验组 ,同龄WKY大鼠作为正常对照组 ,实验组分别予以苯那普利、科素亚及苯那普利 +科素亚联合治疗 12周。实验前及实验开始后 4、8、12周各组测定血压、2 4h尿白蛋白排泄 ;比较实验结束时各组血浆及肾脏组织AngⅡ、TGF - β1水平。结果 :苯那普利、科素亚及苯那普利 +科素亚都可以明显降低SHRs血压、2 4h尿白蛋白排泄 ,但以苯那普利 +科素亚效果最好。 3组中血浆及肾脏组织中的AngⅡ水平在科素亚组升高 ,在科素亚以及苯那普利 +科素亚组下降。 3组中血浆和肾脏组织的TGF - β1水平都有显著降低 ,以合用药组降低的程度最大。结论 :联合应用苯那普利和科素亚 ,可以协同降低自发性高血压大鼠的血压 ,进一步延缓高血压造成的肾脏损害。Objective:To study the protective effect of the combination of benazepril and losartan on renal injury in SHRs.Methods:This study took 13 month SHRs as treated groups,and WKY rats of the same age as control group.The treated groups were fed with benazepril, losartan,benazepril plus losartan respectively for 12 weeks.Blood pressure and 24 hour proteinuria excretion are measured at pre-study and 4, 8, 12 weeks after the study. AngⅡand TGF-β1 level in plasma and kidney were examined in each group when the study was finished.Results:All benazepril, losartan and Benazepril+losartan could lower BP,24 h proteinuria excretion,and the last had the best effect.The AngⅡlevel in plasma and kidney increased in losartan group,decreased in benazepril and combination groups.The TGF-β1 level in plasma and tissue in all the three treated groups decreased, and deceased most remarkably in the combination group.Conclusion:Compared with monotherapy,the combination of benazepril and losartan had additive hypotensive effect,and had better protective effect on renal injury in SHRs.
