己烯雌酚完全抗原的合成和多克隆抗体的制备

摘　　要：采用混合酸酐法 ,将己烯雌酚与牛血清白蛋白或卵清蛋白偶联形成完全抗原。经紫外分光光度计扫描鉴定 ,并通过聚丙烯酰胺电泳实验 ,推算出牛血清白蛋白与DES HS的结合比是 1∶2 5～ 1∶30。以己烯雌酚 -牛血清白蛋白为抗原免疫家兔 ,制备出多克隆抗体 ,经酶联免疫吸附试验和琼脂扩散试验进行鉴定 ,并采用酶联免疫吸附方法进行效价测定 ,抗血清效价均超过了 1∶1 0 0 0 0。The complete antigen, DES-BSA and DES-OVA, were synthesized by combining diethylstilbestrol(DES) with BSA or OVA. The complete antigen was identified by UV spectrametric scanning method. The conjugated ratio of BSA and DES-HS in DES-BSA was 1∶25～1∶30 calculated by polyacrylamide gel electrophoresis. Rabbits was immunized with DES-BSA to obtain the multi-clonal antibody sera.Antibodies were identified by ELISA and agar-gel diffusion experiment. The titer of the antisera by ELISA method were above 1∶10 000.
