摘　　要：应用本室纯化的TsAb免疫的 BALB/C 小鼠,其脾细胞与 SP2/O 骨髓瘤细胞融合,获得了6株分泌抗人 TsAb 单克隆抗体的杂交瘤细胞株,并对其免疫学特性进行了研究。腹水效价为1∶10~6,与人 TG、TM 无免疫交叉反应,抗体亚型为 IgG2a。用辛酸-硫酸铵二步沉淀法对抗体进行了纯化。应用 ELISA 法初步测定了 Graves 氏病患者血清中 TSI 的含量,其阳性率平均为88%。Thyroid stimulating antibody(TsAb)was purified in our lab.and used as antigento immunize BALB/C mice.The splenic cells of the mice were fused with SP 2/0mouse myeloma cells.Six hybridoma cell lines secreting antihuman TsAb were estab-lished.The immunological property of McAb was studied.The ascites titer was 1∶10~6.The McAb did not present a cross reaction with TG and TM.IgG2a subclass wasdetermined by agar double diffusion test.TsAb level was evaluated by ELISA.Theresult showed that average positive rate was found to be 88% in patients with Graves'disease.This method was beleived to be sensitive and simple,and could be used as auseful parameter for diagnosis,therapy and study of pathogenesis of autoimmune thy-roid disorders.
