摘  要:目的:探讨缓激肽β2 受体基因启动子区 91 位至 7 位基因变异与新疆哈萨克族人原发性高血压的关系。方法:运用聚合酶链反应-单链构象多态性分析(PCR SSCP)方法,对 118 例收缩压≥150 mmHg和/或舒张压≥95 mmHg的哈萨克族原发性高血压病人和 65 例年龄、性别、族别、BMI相匹配的血压<135/85 mmHg的正常血压者,分别进行缓激肽β2 受体基因启动子区 91位至 7位基因多态性检测,观察不同SSCP带型在高血压病人组和正常血压对照组中的分布频率。结果:新疆哈萨克族人缓激肽β2 受体基因启动子区存在 A、B、C 3 种带型,A、B、C带型在原发性高血压病人组和正常血压对照组的分布频率分别为 0.28、0.37; 0.22、0.23; 0.50、0.40。3种带型的分布频率在两组间差异无统计学意义(χ2= 2.002, P=0.367)。结论:新疆哈萨克族人缓激肽β2 受体基因启动子区 91位至 7位存在有3种SSCP带型,提示该区域可能存在有基因变异,但该变异可能与哈萨克族人原发性高血压不相关。Objective: To investigate the association between the genetic variants in the promoter region of bradykinin β-2 receptor gene and the essential hypertension in Kazak of Xinjiang. Methods: This study selected 118 hypertensive patients(SBP≥150 mmHg and/or DBP≥95 mmHg)and 65 normotenive controls(BP<135/85 mmHg)matched with sex 、age、BMI from Kazak of Xinjiang. The measurement of the genetic variants was performed by PCR-SSCP in hypertensive patients and normotensive controls respectively. Results: The A、B、C SSCP band patterns were detected in the experiment. The distributed frequencies of the A、B、C SSCP band patterns were 0.28、0.37; 0.22、0.23; 0.50、0.40 respectively in hypertensive group and normotensive group. There was no significant difference between the two groups (χ+2= 2.002,P= 0.367). Conclusion: In Kazak of Xinjiang, there are three different SSCP band patterns in the promoter -91 to -7 region of bradykinin β-2 receptor gene. It suggest thats there exist variants in the promoter region of bradykinin β-2 receptor gene, but the variants may not be associated with essential hypertension.
