摘  要:目的:促愈合药W_11-a_12能显著提高伤口中性粒细胞(neutrophils,Neu)的数量和功能,观察合并全身放射损伤大鼠伤口Neu凋亡率的变化,并观察促伤口愈合药W_11-a_12的作用。方法:实验于1999-05/10在解放军第三军医大学预防医学院全军复合伤研究所完成。将健康成年大鼠随机分为单纯创伤组和6Gy放射复合伤组。6Gy放射复合伤组动物在接受6Gyγ射线全身照射后立即在背部致创伤,伤口局部给予生理盐水或W_11-a_12(每伤口10mg)。从埋入伤口的海绵分离伤口细胞,分别采用TUNEL法、Westemblot及RT-PCR方法检测伤口Neu凋亡率、Neu中Bcl-2蛋白、mRNA含量,透射电镜观察伤口凋亡的Neu。结果:伤后3,6,12,24及48h6Gy放射复合伤组伤口Neu凋亡率变化曲线呈“∧”型,而单纯创伤组呈“∨”型。伤后6,12,24h6Gy放射复合伤组Neu凋亡率分别为(9.90±2.26)%,(12.50±1.83)%和18.16±5.56)%,显(著高于单纯创伤组的(6.44±1.96)%,(2.74±1.22)%和(7.38±2.10)%(t=2.01,2.81,2.52,P<0.05),同时Neu中Bcl-2蛋白及mRNA表达均低于单纯创伤组。W_11-a_12使用组与对照组伤口Neu凋亡率无明显差异。结论:全身电离辐射后伤口Neu凋亡率增加且变化规律与单纯创伤组明显不一致,这与电离辐射引起伤口愈合延缓有关。AIM:W_(11)-a_(12)(a compound) can accelerate the wound healing in irradiated rats a nd increase the number of neutrophils(Neu) in wound.The aim of the present study is to investigate the changes of apoptosis rate of Neu in wound of irradiated r ats and the effects of W_(11)-a_(12) on the changes. METHODS:The experiment was conducted at the State Key Laboratory of Trauma,Bur ns and Combined Injury,Institute of Combined Injury,College of Preventive Medici ne,Third Military Medical University of Chinese PLA from May to October 1999.Hea lth Wistar rats were divided into simple wound group and wound combined with 6 Gy irradiation group randomly.Rats in the later group were wounded after irradia ted with 6 Gy γrays.W_(11)-a_(12) or saline(10 mg for each wound) was given at loca l wound.Wound Neu were collected from sponges which were implanted in incisions of the rats.The apoptosis rate of wound Neu was measured with TUNEL method.The e xpression of Bcl 2 protein and bcl 2 mRNA were explored with Western blot and RT PCR,respectively.The apoptotic Neu were observed under transmission electron microscope. RESULTS:The curve of apoptosis rate of wound Neu to time showed in “∧”shape in the rats of wound combined with 6 Gy irradiation group,while in “∨”shape in rats of simple wound group 3,6,12,24 and 48 hours after wound.The apoptosis rates of wound Neu were (9.90±2.26)%,(12.50±1.83)%and (18.16±5.56)%in the rats of wound combined with 6 Gy irradiation group at the 6,24 and 48 hours aft er wound,significantly higher than those in the rats of simple wound group[(6.44 ±1.96)%,(2.74±1.22)%and (7.38±2.10)%,respectively](t=2.01,2.81 and 2.52,P< 0.05).And the levels of Bcl 2 protein and mRNA in wound Neu were decreased mar kedly in the former group,as compared with those in the latter group.The apoptos is rate of wound Neu was not significantly different between the W_(11)-a_(12) treate d group and the control group.
