摘 要:目的探讨慢性阻塞性肺疾病(COPD)急性加重期大鼠肺组织基质金属蛋白酶活性的变化及通塞颗粒的治疗作用。方法以肺炎克雷白杆菌经鼻腔滴入的方法建立大鼠COPD模型,酶谱法检测大鼠肺组织基质金属蛋白酶。结果稳定模型组、急重模型组、高剂量组、低剂量组、中药对照组、西药对照组92、72、82、62kD均明显高于空白对照组。急重模型组、西药对照组92、72、82、62kD明显高于稳定模型组,高、低剂量组明显低于中药对照组、西药对照组、急重模型组。结论MMP2、MMP9的活性升高参与COPD肺组织基质损伤。通塞颗粒抑制MMP2、MMP9的活性可能是其治疗COPD的主要机制之一。Objective To explore the change of the activity of matrix metalloproteinase (MMP) of lung tissue in the acute exacerbation of COPD rat models and the treatment effect of Tongse granule .Method The acute exacerbation of COPD animal model was developed by drip Klebs-loeffler bacillus from nasal cavity and the MMP of rat lung tissue was detected by zymography method. Results The 92 , 72, 82, 62 kD levels of lung in acute exacerbation of COPD animal model group, stable model group of COPD, high dosage group, low dosage group, Chinese herbal medicine control group, western medicine control group were all higher than those of normal controls. The levels of ~92, 72, 82, 62 kD in acute exacerbation of COPD animal model group and western medicine control group were obviously higher than those of stable model group of COPD .Levels of 92, 72, 82, 62 kD of lung in high dosage group and low dosage group were all obviously lower than those of acute exacerbation of COPD animal model group, Chinese herbal medicine control group and western medicine control group. Conclusions Increase of the MMP-2 and MMP-9 activity was related to lung tissue injury. Tongse Granule could treat COPD by inhibiting the activities of MMP-2 and MMP-9.
