4-硒硫酸酯多糖体外抗瘤作用及机制的研究

摘　　要：目的　研究 4 -硒硫酸酯多糖体外给药的抗瘤作用及可能的机制。方法　采用体外生长抑制实验 ,台盼蓝排染法计数活细胞 ,计算抑制百分率 ;同位素掺入实验 ,观察药物对肿瘤细胞DNA合成的影响 ;流式细胞仪分析其对细胞周期的影响。结果　 4 -硒硫酸酯多糖对肿瘤细胞有直接细胞毒作用 ,可抑制肿瘤细胞DNA的合成 ,使G1期细胞减少 ,停留在S期。结论　 4 -硒硫酸酯多糖体外给药对瘤细胞有明显的抑制作用 ,其机制可能与抑制瘤细胞DNA合成有关。Objective To study inhibitory effects and probable mechanism of Se-carra on tumor cells in vitro.Methods Growing inhibitory test was used in vitro,living cells were counted by trypan-blue staining method,and calculating inhibitory percentage.Effects of Se-carra on DNA synthesis of tumor cells were observed with isotope incorporation experiment and cell cycle was analysed by flow cytometer.Results The experiment showed that Se-carra has direct cytotoxicity on tumor cells,reducing G 1 stage cells,inhibiting G 2+M stage cells,making cells stop Stage,and interfereing DNA synthesis.Conclusion These results indicated that Secarra has obviously depressant effects on tumor cells in vitro.It is probable that these effects relate to inhibiting DNA synthesis.
