摘 要:目的用新西兰兔建立糖尿病性勃起功能障碍(ED)动物模型,为ED的基因治疗提供实验基础.方法雄性新西兰白兔(2.4~2.6)kg30只,实验兔15只采用四氧嘧啶耳缘静脉注射(150mg/kg)诱发糖尿病,对照组兔15只耳缘静脉注射等量生理盐水.在相同条件下饲养,6月后比较试验组兔和对照组兔阴茎勃起功能的改变:方法是测量两组兔基础海绵体内压(ICP)和电刺激盆神经后检测ICP的变化,同时检测颈动脉血压(SAP),鉴定ED兔的建立.结果用四氧嘧啶诱导的糖尿病兔的血糖值明显高于对照组兔,血糖均大于20mmol/L,体重增加小于对照组兔,两组数值有显著性差异.两组兔基础ICP无明显差异,P>0.05;电刺激盆神经时,糖尿病组兔的平均ICP为(11.65±1.40)mmHg;而对照组兔为(33.61±3.20)mmHg,P<0.01,差异有显著性.结论用四氧嘧啶可诱导新西兰雄兔建立糖尿病动物模型,进一步建立ED的动物模型,为研究ED的基因治疗奠定实验基础.Objective To establish a model of diabetic erectile dysfunction (ED) and provide an experimentalfoundation for the gene therapy of ED patients. Methods Half of 30 male New Zealand white rabbits (2.4~2.6kg)(selected at random) were injected intravenously with alloxan (150mg/kg) to make experimental models of diabetesmellitus. The remaining animals were injected intravenously with saline alone. After 6 months, cavernous body pres-sures and arterial pressure were characterized during surgical manipulation and during electrical stimulation of thecavernous nerves. Results Baseline cavernous pressure of normal rabbits was not significantly different with diabeticrabbits. Electrostimulation of pelvic nerve caused ICP to rise to approximately (33.613.20) mmHg in normal rabbits,but (11.651.40) mmHg only in diabetic rabbits. Conclusions This work will provide an experimental foundation forthe gene therapy of ED patients.
