摘  要:目的 探讨小儿病毒感染与微卫星DNA的关系。方法 取经咽拭子病毒分离证实为呼吸道合胞病毒或腺病毒感染的患儿血提取DNA ,用聚合酶连反应对基因座D5S818进行扩增 ,聚丙烯酰胺凝胶电泳分离DNA片段 ,成象系统分析D5S818基因片段长度多态性 ;并分析D5S818基因片段长度多态性与病毒感染临床表现的相关性。结果 发现病毒感染后D5S818基因座等位基因频率与正常对照组有明显差异 ,平均AGAT重复数 ,观察组较对照组略高 ,A GAT重复数 <12和≥ 12两组相比较 ,观察组明显高于对照组。病毒感染组D5S818基因杂合度明显低于对照组。结论 病毒感染可能使DNA微卫星发生改变。Objective Discuss the relevance of virus infection in children and DNA microsatellite. Methods DNA is extracted from peripheral blood of children infected by respiratory syncytial virus or adenovirus by throat swab virus isolation. The DNA is amplified with D5S818 by PCR, analysed with photographic system after polyacrylamide del electrophoresis gel electrophoresis. Discuss D5S818 gene fragment length polymorphism and the relevance of clinic situation of virus infection. Results It is found that after the virus infection, there is a significant difference between the infected group and the control group in D5S818 genotype frequency. The average repetition copy number of AGAT of observe group is a little bit higher than that of the control group,, AGAT copy number<12 and ≥12 of observe group is much higher than that of the control group. The observe group heterozygosity is significant lower than that of the contrast group. Conclusion Virus infection may change the repetition number of the DNA micro satellite.
