摘 要:目的探讨猪软骨细胞同种异体移植的免疫排斥机制。方法分离制备猪软骨细胞和淋巴细胞 ,采用同种异体软骨细胞—淋巴细胞混合培养反应和IFN γ诱导比较分析 ,经3 H TdR法观察淋巴细胞刺激指数 ,FACS测定软骨细胞表面猪白细胞抗原(SLA)分子表达变化 ;不同时间收集混合反应中淋巴细胞 ,分别应用PCR和ELISA技术分析其表达的IFN γ、IL 4mRNA和蛋白含量。结果成功制备了猪的软骨细胞和淋巴细胞 ,软骨细胞表达SLAI类分子 ,而不表达SLAII类分子 ;同种异体软骨细胞对淋巴细胞有轻微刺激扩增作用 ,经IFN γ诱导具有明显增强作用。混合培养 ,18h后上清中可检测到IFN γ蛋白 ,而IL 4表达无变化。结论软骨细胞SLAI类分子刺激同种异体淋巴细胞产生IFN γ,进一步诱导软骨细胞SLAII类分子表达 。Objective To explore the mechanism of immuno-rejection for the swine allogenic chondrocytes in transplantation. Methods The chondrocytes and lymphocytes were isolated from chondrocytic tissues and peripheral blood in the swine. The phenotypes of the chondrocytes were analyzed by FACS and their antigenicity was determined in a swine allogenic chondrocyte-lymphocyte mixed reaction(sCh-MLR) system, in which the lymphocytes of the Banna strain and the chondrocytes of the Xiang-zhu strain were compared as responding and stimulating cells by using 3H-TdR incorporation test, after treatment with swine IFN-γ. The IFN-γ and IL-4 mRNA in the lymphocytes of sCh-MLR were determined, and the culture supernatant of sCh-MLR was assayed for IFN-γand IL-4 proteins by ELISA methods in different times. Results The swine chondrocytes and lymphocytes were isolated successfully. The chondrocytes, which were detected to express the swine leukocyte antigen (SLA) class I molecules constitutively, were not able to initiate a significant allogenic response of lymphocytes. However, when the chondrocytes began to express the SLA Class II molecules after treatment with swine IFN-γ, the lymphocytes were obviously proliferated. IFN-γ protein was detected in culture supernatants of sCh-MLR, but IL-4 was not detected. Conclusion SLA class I on the chondrocytes might stimulate allogenic lymphocytes to produce IFN-γ, in which the inducible expression of SLA class II molecules might be a critical factor attributable to the immuno-rejection reactions.
