摘　　要：目的研究红细胞趋化因子受体(ECKR)在寻常型银屑病患者外周血红细胞(RBCs)中的表达情况,探讨其在银屑病发病机制中的作用。方法应用流式细胞仪和特异性FITC标记的鼠抗人ECKR抗体检测寻常型银屑病患者及正常人外周血RBCs表ECKR蛋白的表达水平。应用RT-PCR方法和ECKR特异性引物检测银屑病患者及正常人外周血红系细胞内ECKRmRNA的表达;并将结果与患者皮损面积及严重程度指数(PASI)进行了相关性分析。结果寻常型银屑病患者外周血RBCs膜ECKR阳性率明显低于正常人,进行期患者较静止期患者显著降低,且银屑病患者RBCs膜ECKR的阳性率与其PASI评分呈负相关。银屑病患者外周血红系细胞内ECKRmRNA的表达水平高于正常人,而进行期患者ECKRmRNA的表达又高于静止期患者。结论ECKR在银屑病发病过程中发挥一定的作用,其表达水平与银屑病患者的病情密切相关。Objective To determine the expression of erythrocyte chemokine receptor (ECKR) on peripheral blood Red blood cells (RBCs) of patients with psoriasis and to explore its clinical significance. Methods ECKR on peripheral blood RBCs was detected by direct immunofluorescence staining method. ECKR on RBCs, which combined with FITC-labeled ECKR mAb, was detected by flow cytometry. Reverse transcription-polymerase chain reaction (RT-PCR) technique was applied to semi-quantatatively analyze ECKR mRNA expression in peripheral blood RBCs from psoriatic patients and healthy controls. The correlation between ECKR and psoriatic aera and severity index (PASI) was evaluated. Results The results showed that the positive rate of ECKR+ RBCs in psoriasis was significantly lower than that in the controls. The positive rate of ECKR+ RBCs in active psoriasis was lower than that of resting psoriasis. Furthermore, the positive rate of ECKR+ RBCs in psoriasis was close related with PASI score of psoriasis. In other hand, ECKR mRNA levels in RBCs from psoriatic patients were markedly higher than those in normal controls. Conclusion The present study suggested that the percentage of ECKR+ RBCs in the patients with psoriasis is significantly lower and is related with the disease progress, ECKR could be regarded as clinical reference index to observe the change of the disease progress.
