摘 要:目的 研究茵栀清肝汤对四氯化碳诱导急性肝损伤大鼠的保护作用。方法 Wistar大鼠随机分为正常组、急性肝损伤模型组、中药治疗低、中、高剂量组 ,中药组分别予以茵栀清肝汤 3g/kg、6g/kg、12g/kg每日一次灌胃 ,共 7d。末次灌胃后除正常组外 ,其余所有大鼠予以 5 0 %四氯化碳 5ml/kg灌胃 1次 ,2 4h后处死全部大鼠 ,收集肝组织及血清标本。对肝组织进行HE染色病理检查 ;生化方法检测大鼠血清天门冬氨酸氨基转移酶 (Aspartatetransaminase,AST)、谷氨酸氨基转移酶 (Alaninetransaminase,ALT)的活性 ;测定大鼠血清超氧化歧化酶 (Superoxidedismutase ,SOD)及丙二醛 (Malondialdehyde ,MDA)的含量。结果 中药茵栀清肝汤治疗组大鼠血清AST、ALT水平较模型组显著降低 ,显著改善肝组织病理。对急性肝损伤大鼠血清SOD ,GSH -PX的活性有明显的升高作用并降低血清MDA的含量。茵栀清肝汤对急性肝损伤大鼠的保护作用呈剂量依赖模式 ,服用 12g/kg剂量大鼠疗效最佳。结论 茵栀清肝汤具有显著保护四氯化碳所致大鼠急性肝损伤的作用。Objective To investigate the effect of Yinzhi Qinggan decoc t ion on the carbon-tetrachloride (CCl 4) induced acute liver injury in rats. Methods Seventy Wistar rats were randomly divided into five groups of 14 rats each: normal group, model group, low dose group, middle dose grou p and high dose group.The treatment groups were orally administered with Yinzhi Qinggan decoction 3, 6 and 12 g/kg per day separately,whereas the normal and mod el groups were orally administered with saline.Except normal rats,all the other rats were administered CCl 4 3.0 ml/kg orally once just after adminstration for of Yinzhi Qinggan decoction seven days.The rats were sacrificed 24 h after CC l 4 administration. Serum and liver samples were collected for analysis. The ef fe ct of Yinzhi Qinggan decoction was determined based on the serum levels of aspar tate transaminase (AST), alanine transaminase (ALT), superoxide dismutase (SOD) and malondialdehyde (MDA). The liver function was examined by biochemistry, and the histopathology of liver was observed by HE staining.Results All rats treated with Yinzhi Qinggan decoction were improved in a dose-depend ent manner up to 12 g/kg, and the maximum effects were observed at the dose of 1 2 g/kg. Serum ALT, AST,and MDA concentrations in untreated rats increased, peake d at 12 g/kg, and decreased gradually in treated rats. Serum SOD concentration i n rats treated with Yinzhi Qinggan decoction was significantly higher than that observed in untreated rats. The liver histological changes were also im proved significantly in the treated groups.Conclusions Yinzhi Qinggan decoction has protective effect on rat acute liver injury induced by CC1 4 administration.It diminishes the severity of liver injury by decr easing oxidative stress due to its possible scavenging effect.
