摘  要:目的用PCR技术检测转人组织蛋白酶K基因鼠系,筛选携带目的基因的阳性小鼠。方法用PCR技术,对显微注射后出生的转人组织蛋白酶K基因小鼠、首建鼠的F1、F2、F3及F2代与野生型ICR小鼠的回交后代进行检测。结果共检测显微注射后出生的小鼠25只,阳性为1只,阳性率为4%;检测F1、F2、F3代小鼠分别为60、77和69只,阳性仔鼠数为35、56和63只,阳性率分别为550%、727%和913%;对回交后代进行检测,未见纯合的F2代小鼠。结论外源基因(人组织蛋白酶K基因)已经整合到小鼠的染色体上,并且按孟德尔遗传定律中的分离定律进行遗传。Objective To detect transgenic mice of human Cathepsin_K gene by using PCR. Methods The founder transgenic mouse and wild ICR mice were intercrossed to obtain F1 generation. The F1, F2, F3 generations and the backcross offspring between F2 and wild ICR were detected by PCR. Results 25 transgenic mice were obtained through microinjection, one was detected to be positive,the positive ratio was 4%;60 F1 genetation, 77 F2 generation and 69 F3 generation were detected. The positive mice and ratio were 35(55 0%) in F1;56(72 7%) in F2; 63(91 3%) in F3, respectively. The backcross offspring of F2 were detected and there wasn’t any homozygote yet in F2 generation. Conclusion The foreign gene (human Cathepsin K gene) has already integrated into mouse chromosome,and it can inherit in accordance with Mendelian inheritance.
