摘  要:目的 研究甲巯咪唑 (MMI)对体外培养人甲状腺细胞功能的影响 ,以探讨MMI的药理作用 ,为指导临床用药提供一个实验依据。方法 人甲状腺细胞培养 2d ,之后分别培养在 5H和 6H液中 ,不同浓度MMI(1、 10、 10 0 μmol/L)状态下孵育 2d。用改良的愈创木酚法测定TPO活性。用放免法测定T3、T4。以竞争性蛋白结合法测定cAMP。结果 在 1~ 10 μmol/LMMI浓度下甲状腺细胞内的TPO活性有较明显的增高 ,而10 0 μmol/LMMI则抑制TPO活性。甲状腺细胞分泌的T3、T4与TPO活性的变化保持一致。MMI在较大范围(1~ 10 0 μmol/L)内不影响细胞内的第二信使cAMP的含量。 结论 MMI对体外培养人甲状腺细胞的功能有影响。Objective To explore MMI pharmacology action, the effects of MMI on human thyroid cells function were studied in culture human thyroid follicles to help clinical therapy and provide a experimental foundation. Methods Thyroid follicles were cultured for 2 days. Then, the follicles were exposed to MMI (1,10,100μmol/L) in 5H or 6H for 2 days. TPO activity was measured by the guaiacol oxidation method. Radio-immunoassay were used to detect T 3,T 4 secretion by thyroid cells and cAMP productions in cultured system.Results Exposure to MMI (1~10μmol/L) caused a significant increased in cellular TPO activity. 100μM MMI inhibited cellular TPO activity. The changes of T 3,T 4 secretion that affected by MMI were consistent with those of TPO. MMI failed to affect cellular cAMP generation over a broad range of concentrations (1~100μmol/L) in cultured cells. Conclusions MMI plays an important role in affecting thyroid cells function in cultured human thyroid follicles.
