高效液相色谱定量测定降压乐的有效成分

摘　　要：降压乐片含去甲氧利血平和甲氯噻嗪两种成分,原料是美国ABBOTT药厂生产的。国外资料提供的含量测定方法是:“用两种不同牌号C_(18)色谱柱,两种流动相,两个内标物,两个检测波长分别测定两种成分。”操作麻烦,费时,成本高,本文利用反相分配色谱法,采用一根C_(18)色谱柱,以甲基炔诺酮为内标物,以甲醇为流动相,在254nm和225nm下分别测定甲氯噻嗪和去甲氧利血平的含量。本文采用的色谱系统:“C_(18)柱理论塔板数依内标物甲基炔诺酮峰计算应不低于一万。内标峰与甲氯噻嗪峰,去甲氧利血平峰分离度应不小于1.7。In this paper a method for separating and determining the contents of the two effective ingredients, deserpidine and metbyclothiazide in Jiangyale tablet by the reversed phase HPLC is presented. In the method, μ-BONDAPAK C13 was used as the column packing; norgestrel as the internal standard and methanol as the mobile phase at the wavelength of 225nm and 254nm. The linear range of deserpidine was 0.7μg-3.0μg (r=0.9999) and recovery was 99:2% (CV:0.6%): The linear range of methyclothiazide was 0.4μg-3.0μg(r=0.9999) and recovery was 99.4% (CV0.6%).
