摘  要:目的:应用离体培养乳鼠胰岛细胞,探讨1,6-二磷酸果糖(fructose-1,6-diphosphate,FDP)对白细胞介素1β(IL-1β)损伤的胰岛细胞是否具有保护作用。方法:实验选用出生1~3d的Wistar大鼠20只。分离培养后的胰岛细胞,将其随机分为对照组、FDP组、IL-1β损伤组、IL-1β+FDP组,每组平行孔为6孔。应用MTT法检测对照组、IL-1β损伤组以及不同浓度(2.5,5.0,7.5,10.0,12.5,15.0mmol/L)FDP保护组胰岛细胞的细胞活性。结果:IL-1β损伤组细胞活性(A值:0.116±0.012)明显低于对照组(0.252±0.020)(F=8.92,P<0.01),加入5~10mmol/LFDP可使细胞活性增强(F=13.35,22.56,P<0.01),但2.5,12.5,15.0mmol/LFDP与受损细胞共同孵育后未能使细胞活性升高(P>0.05)。10,40h时未见FDP对受损细胞活性的保护,20h时IL-1β+FDP组细胞活性(A值:0.219±0.004)明显高于IL-1β损伤组(0.178±0.010)(F=19.10,P<0.01),30hIL-1β+FDP组细胞活性明显高于IL-1β损伤组(F=16.05,P<0.01)。结论:FDP对IL-1β损伤的胰岛细胞活性具有保护作用,且具有时间和剂量依从关系。AIM:To study whether the fructose-1,6-diphosphate(FDP) could protect the pancreatic islet cells injured by interleukin-1 beta (IL-1β). METHODS:Totally 20 Wistar rats,which were born 1-3 days,were chosen.The cultured pancreatic islets cells were isolated,and the cells were divided into control group, FDP groups,IL-1βinjury group,IL-1β+FDP group at random,and each parallel bore was 6 bores.The activities of pancreatic islets cells in the control group,the IL-1βinjury group and the different concentrations(2.5,5.0,7.5,10.0,12.5,15.0 mmol/L)of FDP groups were examined with MTT method. RESULTS:The activity of pancreatic islets cells in IL-1βinjury group (A value:0.116±0.012)was significantly lower than that in control group (0.252±0.020)(F=8.92,P< 0.01).After adding FDP(5-10 mmol/L),the activity of damaged cells were increased(F=13.35,22.56,P< 0.01),but was not increased in the concentrations of 2.5,12.5,15 mmol/L FDP(P >0.05).After adding FDP for 10 hours,40 hours,there was no protection on damaged cells,but after 20 hours,the activity of pancreatic islets cells in IL-1β+FDP group (A value:0.219±0.004) was significantly increased as compared with IL-1βinjury group (0.178±0.010)(F=16.05,P< 0.01),after 30 hours, the pancreatic islets cells activity in IL-1β+FDP group was remarkably increased as compared with IL-1βinjury group(F=19.10,P< 0.001). CONCLUSION:FDP can protect pancreatic islets cells damaged by Il-1βat the dose-and time-dependent manner.
