钙拮抗剂MN9202对心肌肥厚大鼠全血和心肌中5-HT的影响

摘　　要：目的 :研究二氢吡啶类钙拮抗剂MN92 0 2对心肌肥厚大鼠全血和心肌组织中 5 HT的影响 .方法 :在L 甲状腺素诱发的大鼠心肌肥厚模型上 ,用荧光分光光度法测定心肌肥厚大鼠全血和心肌组织中 5 HT含量 ,免疫组化法测定心肌组织中 5 HT受体相对含量 ,并观察MN92 0 2对上述指标的影响 .结果 :模型大鼠全血和心肌组织中 5 HT含量明显升高 (1.3± 0 .3vs 2 .1± 0 .2mg·L -1,2 99± 5 7vs 4 2 8± 5 8ng·g-1,P <0 .0 5 ) ,心肌组织中 5 HT受体相对含量增加(0 .30 0± 0 .0 12vs 0 .180± 0 .0 16 ,P <0 .0 1) .给予两种剂量的MN92 0 2 ,能够使全血和心肌中 5 HT减少 (P <0 .0 5 ) ,心肌组织 5 HT受体相对含量降低 (P <0 .0 1) .结论 :MN92 0 2可能通过阻止 5 HT的合成或 /和减少 5 HT的释放或影响 5 HT受体 ,从而影响心肌肥厚的发生发展 ,这可能是MN92 0 2防治心肌肥厚的机制之一 .AIM: To study the effects of methylpentyl1, 4 dihydro 2, 6 dimethyl 4 (3 nitrophenyl ) 3, 5 pyridinedicarboxylate (MN9202), a novel DHPs calcium antagonist, on the 5 HT contents in blood and in myocardium in rats with cardiac hypertrophy. METHODS: The contents of 5 HT in blood and myocardium and the relative amount of 5 HT receptor (5 HTR) in cardiac tissue were measured by fluorescent spectropbotometry and immunohistochemistry. The effects of MN9202 on cardiac hypertrophy induced by L thyroxine were investigated. RESULTS: The contents of 5 HT in blood and myocardium in rats with cardiac hypertrophy were significantly increased (1.3±0.3 vs 2.1±0.2 mg·L -1 , 299±57 vs 428±58 ng·g -1 , P <0.05). The relative amount of 5 HTR positive cells in cardiac tissue also increased obviously(0.300±0.012 vs 0.180±0.016, P < 0.01). The contents of 5 HT in blood and in myocadium were decreased ( P <0.05 ) and the relative amount of 5 HTR in cardiac tissue were also decreased ( P <0.01) when rats were pretreated with MN9202 at different doses. CONCLUSION: MN9202 may play a role in blocking 5 HT synthesis and reducing its release, or affecting the 5 HTR, which may account for MN9202 in preventing myocardial hypertrophy.
