多头绒泡菌G_2期cDNA表达文库的构建

摘　　要：cDNA表达文库是分离、筛选基因的重要平台.为分离多头绒泡菌SC35类似蛋白PSCL32 5及其激酶的基因,首先检测多头绒泡菌PSCL32 5及其激酶在细胞周期不同时相的含量,发现PSCL32 5在G2期含量最高;再从多头绒泡菌G2期RNA出发,构建多头绒泡菌G2期cDNA表达文库.得到cDNA文库的初始滴度为1 22×106pfu/mL(pfu为噬菌体数目),cDNA扩增文库的滴度为1 12×1011pfu/mL,插入片段的重组率接近100%,插入片段的大小在0 5～2 5kb范围,说明所构建的cDNA文库质量较好.In order to isolate the genes of SC35-like proteins and SRPK-like proteins of Physarum polycephalum, a cDNA expression library of P. polycephalum at G_2 phase was constructed. The titer of the primary cDNA library is 1.22×10~6 pfu/mL and of the amplified library is 1.12×10^(11) pfu/mL(plaque-forming units,pfu). Almost all of the phages in the cDNA library were reconstituted. The size of the inserts ranges from 0.5 kb to 2.5 kb. All of the above mentioned have answered for the general requirements of a cDNA expression library.
