摘 要:目的探讨羟苯磺酸钙(calcium dobesilate)对血肌酐的影响及可能机制。方法诱导大鼠2型糖尿病后,随机分为糖尿病对照组(C组)及羟苯磺酸钙治疗组(D组),24周后比较各组大鼠的血肌酐(Scr)、尿素氮(BUN)、蛋白激酶(PKC)、转化生长因子(TGF-β1)、IV型胶原(type IV Collagen)及血管紧张素II(AgII)水平,用光镜、电镜观察肾脏组织,用质子谱分析仪分析羟苯磺酸钙与肌酐结合情况。结果羟苯磺酸钙治疗组血肌酐水平较糖尿病对照组明显下降,肾脏PKC,TGF-β1,type IV Collagen水平,D组较C组减少,D组和C组间的AgII和Ccr水平无差异,光镜、电镜示D组病理改变较C组明显改善,质子谱分析仪分析证实羟苯磺酸钙可以和肌酐牢固结合。结论羟苯磺酸钙通过化学结合及改善肾脏病理结构来降低血肌酐水平。Objective: To investigate the effects and mechanisms of calcium dobesilate on serum creatinine(Scr). Methods: Type 2 diabetic rats were randomly divided into control group ( group C) and experimental group ( group D) treated with calcium dobesilate. The serum creatinine (Scr), Protein Kinase C (PKC), angiotensin II (AgII), Creatinine clearance (Ccr), transforming growth factor-beta1 were compared among the groups after 24 weeks. The renal tissue were observed by light microscopy and electron microscopy. Conjugation of Scr and calcium dobesilate was analyzed through mass spectrographic. Results: After 24 weeks, PKC, TGF-β1, Collagen IV, Scr, BUN in group D were significantly lower than the group C, meanwhile, renal pathologic changes in group D improved. Ccr and AgII had no difference between group C and group D. Mass spectrographic analysis proved that the calcium dobesilate can conjugate creatinine. Conclusion: Calcium dobesilate can decrease Scr through ameliorating renal lesions and conjugating creatinine.
