摘 要:目的制备羟基喜树碱纳米乳注射液,初步研究其抗癌作用。方法运用相变温度法制备羟基喜树碱纳米乳注射液,以MTT法测定羟基喜树碱纳米乳注射液与羟基喜树碱注射液对小鼠S180癌细胞的抑制作用,并比较了它们对小鼠移植瘤Heps抑制作用。结果羟基喜树碱纳米乳注射液与羟基喜树碱注射液对小鼠S180癌细胞的IC50分别为131和455μmol/L;与羟基喜树碱注射液组相比,羟基喜树碱纳米乳注射液抑制Heps的肿瘤生长作用可增加145%(P<005)。结论羟基喜树碱制备成纳米乳注射液后大大提高了其抑制癌细胞的能力。AIM:To develop o/w nano-emulsions as potential 10-hydroxycamptothecin (HCPT) delivery systems and to investigate their antitumor effects.METHOD: The incorporation of HCPT into the nano-emulsions has been done by the Phase Inversion Temperature (PIT) method.The in vitro antiprolifeative activity on cultured S_~180 cells was evaluated by the MTT method.The in vivo antitumor effects were studied in mice bearing Heps tumors. RESULT: The average diameter and zeta potential of HCPT nano-emulsions were 46.6 nm and-44.1 mV.The HCPT nano-emulsions showed more activity against S_~180 cells than HCPT injections,with IC_~50 values of 1.31 and 45.5μmol/L respectively.CONCLUSION:The HCPT nano-emulsions have higher antiumor effects than HCPT injections, and they are promising antitumor drug delivery systems.
