摘 要:目的 建立丹参中丹酚酸B的优化提取条件,并确立其HPLC测定方法。方法 丹参粉末在适当的溶剂中冰浴超声,提取物进行HPLC分析。色谱柱为Alltlmac18(5μm,250mm×4.6mm);流动相为乙腈-0.5%冰醋酸水溶液,采用梯度洗脱程序,流速为1.0mL/min;紫外检测波长为290nm。结果 在冰浴超声条件下,70%甲醇对丹酚酸B提取率最高。水的提取率为70%甲醇的95.06%。丹酚酸B在0.07~0.70μg范围内呈良好的线性关系(r=0.9997)。精密度实验中RSD为1.62%,稳定性和重现性实验中RSD均小于3%。结论 本实验提取丹酚酸B处理方法简单,测定结果稳定可靠,可作为丹酚酸B含量测定方法。Objective To optimize the conditions for extracting Salvianolic acid B from Danshen (Salvia miltiorrhiza Bge. ). Methods Danshen crude drugs were dried, ground and sieved through 60-mesh screen sieve, followed by 20min ultrasonication in ice bath with different solvents. The Danshen suspensions were filtered and Salvianolic acid B in the filtrate was determined by HPLC. The column used was Alltima C18 (5μm, 250 mm×4. 6 mm); The mobile phase was with acetonitrile-0. 5% acetic acid solution in program of elution; the flow rate was 1. 0 mL/min and the detect wavelength was 290 nm. Results With ice bath, 70% methol achieved the highest rate of extraction, and with-water extraction was 95. 06% of that with 70% methol. Repeatability and precision were good. Salvianolic acid B had a good linearity within the range of 0. 07-0. 70μg (r=0. 9997),and the RSD was less than 3%. Conclusion The treatment of samples was simple and the results were accurate. This method may be used for extraction and determination of Salvianolic acid B.
