摘  要:目的 通过肌内注射弓形虫DNA疫苗PCDNA3.1 P30 P2 2直接免疫小鼠 ,观察其诱导的体液免疫应答及保护性作用。方法 大量制备重组真核表达质粒PCDNA3.1 P30 P2 2 ,肌内注射免疫小鼠 ,ELISA法测定其抗体滴度的变化 ,用弓形虫作致死性攻击感染。结果 实验组抗体水平明显高于对照组 ,二者存在显著性差异 (P <0 .0 5 ) ;攻击感染后实验组小鼠存活时间明显延长 (P <0 .0 5 )。结论 重组真核表达质粒PCDNA3.1 P30 P2 2肌注BALB/C系小鼠可诱导一定的体液免疫应答 ,对攻击感染具有一定的保护性。Objective To directly immunize mice via intramuscular injection with DNA vaccines pc DNA3.1-P30-P22 and to observe the humoral and the protective immune responses.Methods Large-scale prepared recombinant plasmid pcDNA3.1-P30-P22 was injected into the BALB/c mice via muscles to detect IgG antibodies by ELISA.All mice were challenged with highly virulent tachyzoites.Results The level of the IgG antibodies was remarkably higher than that of the control group,and the survival time of immunized mice was longer than that of the control group.Conclusion The recombinant plasmid pcDNA3.1-P30-P22 could elicit BALB/c mice to generate humosal and protective immune responses for Toxoplasma gondii.
