摘  要:目的 :观察滋阴中药知母中活性成分ZMS的不同剂量和不同疗程对老龄小鼠脑M受体的调整作用。方法 :纯系老年Balb c小鼠 ,每天胃管灌服一定剂量ZMS ,以对M受体无选择性3 H QNB为放射配基 ,用单点放射配基结合分析法测定脑M受体密度 ,和灌服溶媒的平行老年小鼠做统计比较。结果 :ZMS 0 5 5 0mg d 2个月后能使老年小鼠脑M受体密度明显高于不服药的同龄对照鼠。ZMS 0 5mg d剂量下 ,随用药时间延长 ,对老龄小鼠脑M受体的上调作用越来越明显 ,用药6 0d和 16 0d组与对照组比较有显著差异。上述大、中、小三种剂量组的死亡率与老年对照组做x2检验 ,均明显降低。结论 :知母甙元ZMS对老龄小鼠脑M受体具有上调作用 ,并与剂量和疗程成正相关。Objective To evaluative the regulation effect of different doses and different time courses of ZMS,the active component from traditional Chinese medicinal herb Zhimu,on brain M receptors in aged mice.Methods The densities of brain M receptors were detected with single point radioligand binding assay with 3H QNB.Aged Balb/c mice were administered with ZMS in CMC Na via stomach tube once daily.The data were compared with parallel aged control mice administered with CMC Na alone.Results The experiment of different doses showed that the M receptor density was significantly increased in aged mice treated with ZMS at doses of 0 5~5 0mg/d/mouse but not at doses of 0 05mg/d/mouse for 2 months when compared with untreated aged mice.Experiment of time course showed that the longer the treatment,the more elevation of brain M receptor density was achieved.There was significant elevation in the group treated with 0 5mg/d/mouse of ZMS for 60 days,and more elevation was obtained in the 160 days group. χ 2 test showed that there was significant decrease of the fatality of mice in the three groups treated with 5 0,0 5 and 0 05mg/d/mouse of ZMS for 60 days than the control aged mice.Conclusions The study demonstrated that ZMS could up regulate M receptor densities in aged mice.This up regulation showed positive correlation to the dose and time course of ZMS treatment.
