ESN、AES在IGSS试验中作为dsDNA底物特异性的进一步研究

摘　　要：对酶处理人精子头核(ESN)、酸洗脱鼠肝、胃印片(AES-肝/肾)在免疫金银染色(IGSS)试验中作为双链DNA(dsDNA)底物的特异性作了进一步研究。结果显示,血清经组白蛋、RNA吸收,底物经加热、胰/糜蛋白酶及RNA酶处理,对检测结果影响不大;而血清经DNA吸收及底物经DNA酶处理,对检测结果影响极大;ESN/AES-IGSS检测结果与并行对照的抗RNA抗体检测结果差异有显著性,其滴度间亦无直线相关性。这表明底物中主要是dsDNA与血清中相应抗体发生反应。抗精子抗体阳性血清及输精管结扎后血清亦均不能使ESN呈阳性反应。因而ESN、AES作为dsDNA底物的特异性应予肯定。A further study on the specificities of enzyme-treated human spermatozoa nuclei (ESN) and acid-elutiou-treated substrates of mouse liver/kidney ( AES-liver/kidney ) as dsDNA substrates in immunogold-silver staining (IGSS) for anti-dsDNA antibodies ( AdsDNA ) was performed. It was found that the results of detection changed when the sera had been absorbed with DNA or when the substrates treated with deoxyribonuclease I. However, the results were not obviously changed when the sera had been absorbed with histone or RNA, or when the substrates heated or treated with trypsin, chymotrypsin or ribonuclease. These data suggest that it is dsDNA in the substrates that reacted to the relevant serum antibody ( AdsDNA).It was also found that the anti-spermatozoa positive serum and the serum from vasoligated man showed no positive reaction to ESN either . So the specificities of ESN and AES as dsDNA substrates in IGSS are definitely good.
