摘  要:阿昔洛韦(又称无环鸟苷,化学名:乙氨基-1,9-二氧-9-(2-羟乙氧基)甲基-6H-鸟嘌呤-6-酮)为核苷类抗病毒药。该药能被病毒的胸腺嘧啶核苷激酶选择性地磷酸化,从而转化为单磷酸酯,后者受细胞酶的作用,再转化为二磷酸酯和三磷酸酯,阿昔洛韦三磷酸可干扰病毒的DNA聚合酶,特别能干扰疱疹病毒的DNA聚合酶,从而抑制病毒DNA的复制。本文建立了以液—固萃取为前处理、反相高效液相色谱法、紫外检测测定人血浆中阿昔洛韦浓度的方法,比较了两种阿昔洛韦片剂的生物利用度。A RP-HPLC method was developed for the determination of aeyclovir (ACV) in human plasma. ACV was extracted from plasma using ODS, obtained from Sep-Pak C_(18) cartridge (Waters associates) and reinstalled in a self-made glass tube suitable for extraction of solutes from small volume plasma samples. After extraction, and rinsing the ODS extract with water to remove hydrophilic impurities, ACV was recovered with methanol and chromatographed on YWG C_(18)H_(37) column, and detected at 254 mm. The mobile phase was 5.0% (v/v) methanol/water. The average recovery of ACV was 73. 5%; the minimum detection concentration of ACV in plasma was 20 ng/ml with a plasma volume of 0. 2 ml and S/N value of 2. A good linear relationship between the peak area ratios and ACV concentrations was found at the ACV concentrations ranging from 0. 2 to 1.0 μg/ml, C_(ACV)=0. 840 A_(ACV)/A_(is)+0.002, r=0.9969.
