凋亡相关蛋白Survivin及Caspase3在肾细胞癌中的表达及其与凋亡的关系

摘　　要：　目的　探讨凋亡相关蛋白Survivin及Caspase 3在肾细胞癌表达及其与肾细胞癌凋亡的关系。方法　应用SP免疫组织化学法检测60例肾细胞癌石蜡切片中 Survivin和 Caspase 3 表达的情况, 并用原位末端标记法 (in situend labeling, ISEL) 标记凋亡细胞, 结合临床资料进行分析。结果　Survivin 在肾细胞癌标本中的表达阳性率为88. 3% (53/60), 而正常对照组中无一例呈阳性表达; Caspase 3在肾细胞癌标本中的表达阳性率为 81 .6% (49/60),与对照组相比差异无显著性意义。Survivin 的表达与肾细胞癌的组织学分级和临床病理分期均显著相关 (均 P<0 .01), 但与组织学分型无关。Caspase 3的表达与肾细胞癌的组织学分级、临床分期及组织学分型均无关。肾细胞癌中Survivin与Caspase 3的表达没有相关性。Survivin阳性组凋亡指数明显低于阴性组 (P<0. 01)。结论　Survivin的表达与肾细胞癌的分化程度和临床病理分期密切相关, Caspase 3蛋白的表达情况与肾细胞癌关系不密切。Survivin 能显著抑制癌细胞的凋亡, 但不能抑制Caspase 3的表达。Objective To investigate the expression of Survivin and Caspase 3 in renal cell carcinoma (RCC) and their relation with apoptosis. Methods The expression of Survivin and Caspase 3 was detected by streptavidin-peroxidase immunohistochemical method in the paraffin-embedded sections of 60 renal cell carcinoma patients with the clinical data analyzed. Tumor cell apoptosis was examined by in situ end-labeling (ISEL) method. Results The positive rate of the expression of Survivin in RCC was 88.3 % (53/60), while none of 10 cases in control group expressed Survivin. The positive rate of the expression of Caspase 3 in RCC was 81.6 % (49/60), but there was no significant difference between the tumor group and the control group. The expression of Survivin was related to the histological grades and the pathological stages, while the expression of Caspase 3 was not so. There was no relation between the expression of Survivin and Caspase 3.Conclusion The expression of Survivin in RCC was associated with the histological grades and the pathological stages. Survivin obviously inhibited the apoptosis of RCC cells, but could not inhibit the expression of Caspase 3.
