摘　　要：目的 :研究新型免疫抑制剂FTY72 0对大鼠肾脏缺血再灌注损伤 (IRI)的预防作用。方法 :制备大鼠肾脏IRI模型 ,从下腔静脉注入不同剂量的FTY72 0 ,观察术后第 1、2、3、5、7d血清肌酐值 (Scr)和术后第 2、7d外周血淋巴细胞数 (PLC)的变化 ,并在术后第 2d取肾脏作组织学检查观察急性肾小管坏死的情况。结果 :FTY72 0处理组动物术后Scr水平显著低于对照组并呈剂量依赖性 ;FTY72 0处理组术后第 2d的PLC显著低于对照组 ;组织学检查显示FTY72 0处理组肾脏的缺血性损伤轻于对照组。结论 :FTY72 0可以减少PLC 。Objective:To study the effect of FTY720 on prevention of ischemia-reperfusion injury in rat kidneys.Methods:Ischemia-reperfusion injury model in rat kidney was established and FTY720 at different dose was injected into the inferior vena cava. Changes in serum creatinine was monitored 1, 2, 3, 5 and 7 day after operation and peripheral blood lymphocyte count was measured on day 2 and 7.Kidneys were fixed for histological analysis of the severity of acute tubular necrosis. Results:The groups treated with FTY720 showed lower postoperative creatinine level dose-dependently compared with the control group. FTY720-treated groups also showed less PLC than the control group. The histological analysis showed that the FTY720-treated groups had less histological change of ischemic injury than that of control group.Conclusion:FTY720 can decrease PLC and prevent ischemia-reperfusion injury in rat kidneys.
