摘  要:目的阐明盐藻水提物(WEDS)对小鼠细胞免疫功能的调节作用。方法以DNCB诱导小鼠迟发性超敏反应(DTH),80和250mg·kg-1环磷酰胺(Cy)分别诱导小鼠DTH反应增强和减弱模型,观察不同剂量的WEDS对小鼠DTH反应的影响;以0.6%巯基乙醇酸钠诱生小鼠腹腔巨噬细胞,观察不同剂量的WEDS对小鼠腹腔巨噬细胞生成IL1的影响;以ConA诱导T淋巴细胞增殖,观察不同浓度的WEDS体外对T淋巴细胞增殖的影响。以ConA诱导T淋巴细胞增殖,观察WEDS不同剂量、不同作用时间体内对T淋巴细胞增殖和IL2生成的影响。结果剂量为100,200和300mg·kg-1·d-1的WEDS对Cy诱导的小鼠免疫功能的增强和减弱均有显著的下调和提升作用;浓度为125~500μg·mL-1的WEDS对腹腔巨噬细胞分泌IL1有显著的促进作用,超出上述范围之外浓度的WEDS对腹腔巨噬细胞分泌IL1的促进作用显著减弱;浓度为62.5μg·mL-1的WEDS体外对T淋巴细胞增殖有显著的促进作用;剂量为100,200和300mg·kg-1·d-1的WEDS能显著促进ConA诱导的小鼠T淋巴细胞增殖、IL2的分泌,其中200mg·kg-1·d-1的促进作用最为显著。结论WEDS对小鼠的细胞免疫功能具有双向调节作用。AIM: To elucidate the modulatory effects on cell immunity of mice by water extract of Dunaliella salina (WEDS). METHODS: DTH of mice was induced by 50% DNCB. The enhanced and suppressed DTH of mice was developed by cyclophosphamide (Cy) at a dose of 80 and 250 mg·kg -1 for observation on effects to DTH by different doses of WEDS. Peritoneal macrophages of mice were induced by 0.6% sodium thioglycolic acid for the assay of impact on interleukin 1 release by different concentration of WEDS. The spleen lymphocytes of mice were used for analysis of in vitro effects on proliferation by different concentration of WEDS. The lymphocytes from spleen and thymus of mice were utilized for examination of in vivo effects on proliferation, the release of interleukin 2 by different doses of WEDS at different duration of treatment. RESULTS:WEDS ig administered at doses of 100, 200 and 300 mg·kg -1 ·d -1 resulted in significant enhancement to DTH in immuno suppressed mice ( P < 0.01 ) and obvious reduction to DTH in immuno enhanced mice ( P < 0.01 ). The promotion on release of IL 1 by WEDS peaked at 125 ~250 μg·ml -1 . Other than the range of concentration, WEDS induced attenuation on enhancement of IL 1 production. In vitro enhancement on proliferation of lymphocytes by WEDS was detected to be significant at 62.51 μg·ml -1 ; while in vivo enhancement on proliferation of lymphocytes by WEDS was assayed to be prominent at a dose of 200 mg·kg -1 ·d -1 and indicated duration dependent manner. WEDS ig treated at doses of 100, 200 and 300mg·kg -1 ·d -1 exhibited duration and dose dependent reinforcement on release of IL 2. Among the designed doses, 200mg·kg -1 ·d -1 was found to be the most effective. CONCLUSION:Adequate dose of WEDS acted not only as enhancing but modulatory agent in cell immunity of mice.
