大鼠抗Thy-1系膜增生性肾炎TGF-β1和calponin h1表达变化的初步研究

摘　　要：目的 探讨转化生长因子-β1(TGF-β1)和碱性调宁蛋白(calponin h1)在大鼠抗Thy-1系膜增生性肾炎 中的表达及其意义。方法 制备大鼠抗Thy-1系膜增生性肾肾炎模型,提取其肾组织的总蛋白和总RNA。应 用免疫组织化学ABC法、Western blot、RT-PCR等方法分别从蛋白和核酸水平检测TGF-β1和calponin h1在肾 炎模型1、3、5、7、14、21、28 d组织中的表达,研究其相互关系。结果 大鼠抗Thy-1系膜增生性肾炎中TGF-β1 的蛋白表达从7天组起明显随时间延长增强,至21 d组达高峰后回落,核酸表达差异不明显;calponin h1的少量 表达见于正常对照组,1、3、5 d组起表达明显增强,7 d组达到最高峰后开始回落,核酸表达的变化趋势则与 TGF-β1的蛋白表达变化趋势一致。结论 大鼠抗Thy-1系膜增生性肾炎组织中calponin h1表达增强,并在肾 炎病变早期呈增强趋势,后者与TGF-β1在肾炎早期的增强表达趋势相似。calponin h1可能在肾炎早期发挥抗 细胞增殖的作用。Purpose To study the relationship and its significance between TGF-β1 and calponin h1 expression in rat anti-thy-1 glomerulonephritis. Methods Anti-Thy-1 glomerulonephritis model was produced in rats by injecting anti-Thy-1 polyclonal antibody. Renal samples were obtained from sacrificed rats in 1,3,5,7,14,21,28 days respectively after injection. Immunohitochemistry, Western blot, RT-PCR were used to detect the expression of TGF-β1 and calponin h1 at protein and nucleic acid level. Results Expression of TGF-β1 increased from 7 day to 21 day at protein level and so did calponin h1 at nucleic acid level. The higest level of calponin h1 protein was found at 7 day. No apparent difference was founded in the expression of TGF-β1 mRNA.Conclusions Elevated expression of TGF-β1 and calponin h1 occured in rat anti-thy-1 glomerulonephritis model. Calponin h1 may play a role in suppression of cell proliferation in early stage of glomerulonephritis.
