摘 要:目的 :分析比较 4株临床分离的淋球菌和 2株标准菌株外膜蛋白PI基因及蛋白质序列 ,初步探索我国淋球菌的变异情况 ,为淋球菌多态性研究、PI基因的表达和疫苗研制提供依据。方法 :构建pBS T PI克隆重组子并进行测序 ,用blastn和CLUSTALW分析基因和蛋白质序列的相似性 ,并与已发表的序列进行比较 ,预测并分析所得PI基因的loop (表面暴露区域 )结构。结果 :在GenBank中搜索到相似性大于 98%的序列 ,6条序列分别属于PIA和PIB两个亚型 ,其中PI 4的序列与GenBank中的相应序列有 18个位点的差异 ,可能为我国特有的流行株。位于 7个loop结构中的变异位点占 3条PIA蛋白间总变异的 6 7% ,占 3条PIB蛋白间总变异的 84 % ,变异程度较高。结论 :成功克隆了临床分离的淋球菌株和标准菌株的PI基因并进行序列分析 ,为深入研究我国流行的淋球菌菌株的多态性、PI基因的表达和疫苗研制奠定了基础。Objective:To analyze PI gene and protein sequences of four clinical isolates and two standard strains of Neisseria gonorrhoea, and primarily study the variability of Neisseria gonorrhoeae in China. Methods:pBS-T-NG recombinants were constructed and sequenced. The sequences were analyzed by using blastn and CLUSTAL W algorithm. Compared with previous researches, the surface-exposed regions (loops) of PIs were predicted.Results:Sequences with homologous rates higher than 98% were found in GenBank database. According to the result of multiple alignments with CLUSTAL W algorithm, PI genes in this study were fall into two subtypes, PIA and PIB. NG-4 most likely be a unique prevalent strain of China, but more studies should be done to validate it in the future. Seven loops were predicted, and proved to be the most various regions.Conclusion:pBS-T-NG recombinants were successfully constructed, sequenced and analyzed. This study will be very helpful for the coming researches on Neisseria gonorrhoeae diversity and vaccine development.
