摘 要:目的 研究刀豆素A(ConA)、结核特异性转移因子(PSm·tb-TF)联用对结核菌感染小鼠的保护效应及作用机制。方法 小鼠感染结核分枝杆菌后1天起,分别给予CbnA和/或Psm·tb-TF治疗,观察各组治疗后半数死亡时间、一定时间内的死亡率、脾脏内结核活菌数;检测脾细胞分泌γ干扰素(IFN-γ)、IL-4及腹腔巨噬细胞(P-Mφ)产生NO水平。结果与感染对照组、ConA单用组、PSm·tb-TF单用组比较,ConA联合PSm·tb-TF组小鼠半数死亡时间明显延长,28天内的死亡率明显降低,脾脏内结核活菌数明显减少。脾细胞分泌IFN-γ水平明显升高;IL-4水平降低。P-Mφ产生NO显著增加。结论 ConA、PSm·tb-TF联合应用对结核菌感染小鼠的保护效应明显好于单用ConA或PSm·tb-TF组。Objective To study the protective effect and mechanism of ConA combined with PSm·tb-TF on mycobacterium tuberculosis infected mice. Methods The mice were treated with ConA and/or PSm·tb-TF by intraperitoneal injection 1 day postinfection challenged with H_(37) Rv by their tail veins. Time of 50% death (T_(50)) and mortality rate within 28 days were investigated. The numbers of viable bacteria in the spleen were counted. The level of 1FN-γ, IL-4 in spleen cells and the level of NO in macrophage cultural supernatant was measured. Results The combination of ConA with PSm·tb-TF prolonged T_(50), decreased mortality rate within 28 days, diminished the number of viable bacteria in the spleen, increased the level of IFN-'/and reduced the level of IL-4 secreted by spleen cells, enhanced the production of NO secreted by macrophages. There was significant difference as compared with non-treated infection group of treated with ConA or treated with PSm·tb-TF alone. Conclusion The protective effect of the combination of ConA with PSm·tb-TF on mycobacterium tuberculosis infected was significantly better than the ConA or PSm·tb-TF used alone.
