摘  要:目的 建立一种敏感而特异的双抗体夹心法检测血液中循环免疫复合物(CIC),并探讨血液中肺炎衣原体(Cpn)CIC与冠心病的关系。方法 包被已知特异性鼠抗人CpnTW183IgG,加入用7%PEG6000,从冠心病患者血清中提取的CpnCIC,再加入酶标二抗与CIC中的抗体结合,通过底物显色检测患者CpnCIC阳性率。结果 冠心病组CpnCIC的阳性率(63.8%)高于对照组(40.0%),差异有显著性意义(X^2=7.78,P<0.05)。结论 建立的双抗体夹心法检测CpnCIC有较高的敏感性和特异性,可用于批量临床标本的检测,血清中中分子量CpnCIC的存在与冠心病之间有密切关系。Objective To establish a sensitive and specific method of double mono-cloned antibody for detection of serum circular immune complex (CIC) and further investigate the relationship between CIC of Chlamydia pneumoniae and the coronary heart disease. Methods The specific mouse anti-human CpnTW183IgG was coated. CpnCIC extracted from the blood of patients with coronary heart disease with 7% PEG6000 was added. Enzyme-labeled second antibody was added to bind with the antibody of CIC. The positive rate of CpnCIC in the patients was measured by the reaction of substrate. Results The positive rate of CpnCIC in the experimental group was higher than in the control group (63.8% vs 40.0%, P<0.05). Conclusion It is a sensitive and specific method of the double layer antibody for detection of CpnCIC. Serum CpnCIC is correlated with coronary heart disease.
