摘 要:对影响花粉超低温保存效果的因素进行了研究,结果表明在—25℃下进行低温锻炼可提高在液态氮(—196℃,LN)中冰冻保存花粉的生存率;负压环境对花粉的超低温保存有积极作用;自来水冲洗是液氮保存番茄花粉的较佳解冻方法;在液氮温度下,没有观察到花粉游离脯氨酸(Pro)含量的变化;液氮中保存的番茄花粉能正常授粉结实,并能产生种子。This paper discusses some factors that affect the viability of tomato (Lycopersicon esculentum Mill. ) pollen stored in Liquid Nitrogen (LN) in order to improve cryopreser-vation effect. The experimental results indicated that pre-freezing at -25 degrees centigrade can increase the viability of pollen stored in LN (table 1 ). The effectiveness of vacuum environment that is 250 mmHg acted positively in tomato pollen cryopreser-vation (table 2 ). Experiment for comparison of 4 thawing methods were carried out. The results showed that the effectiveness of thawing with tap water is best. The free Pro content of pollen stored in LN almost did not vary. The tomato pollen stored in LN for 15 days can pollinate, therefore the flowers pollinated develop fruit and seed.
