SCREENING OF SALT-TOLERANT WHEAT VARIANTS VIA ANTHER CULTURE IN SALT STRESS MEDIA

摘  要:“京花1号”小麦的花药培养在含不同 NaCl浓度,附加 2mg/L 2,4-D和0.5mg/L KT的N_6培养基上,确定筛选浓度。0.3%、0.4%和0.5% NaCl逆境下,接种经γ射线辐照的和未辐照的“京花1号”小麦花药,诱导愈伤组织,并在同样NaCl逆境下分化,获得6株花粉植株。其中经γ辐照,0.5%NaCl下获得的1株,得到1粒种子。经无盐条件下繁殖后,γH_3(照射第3代)种植在中等盐渍地,在0.29%含盐量土壤中生长正常。γH_4代按单株作进一步盐渍地、盐池和水培鉴定,有3个株行其耐盐力明显高于亲本“京花1号”。Anthers of 'Jinghua No. 1' winter wheat were cultured on N_6 basal medium supplemen- ted with 2,4-D 2 mg/L, KT 0.5 mg/L and NaCl of various concentrations for defining selec- tion stress. Irradiated and non-irradiated anthers were cultured on the medium salinized with 0.3%, 0.4% or 0.5% NaCl for callus induction. Then screened calli for differentiation were transferred to medium containing NaCl with the same concentrations as that for callus induc- tion. 6 pollen plants were selected in which 1 pollen plant was obtained from irradiated an- thers on the medium containing 0.5% NaCl. Only one seed was collected from this pollen plant. Growth status in the γH_3 of the above-screened pollen plant was normal in the modera- tely salt-affected soil. It was demonstrated that sal-tolerance of 3 strains in the γH_4 upon fur- ther valuation of water culture, salt-soil tank and field plotting was significantly higher than that of parent.
