甜菜坏死黄脉病毒外壳蛋白基因在大肠杆菌中的高效表达

摘　　要：将甜菜坏死黄脉病毒(BNYVV)内蒙古分离物的外壳蛋白基因亚克隆到pJW2上构建成在大肠杆菌中表达的载体。SDS-PAGE及Westexn blotting检测的结果表明,该表达载体在大肠杆菌DH5 α中经温度诱导后特异地表达21kD的甜菜坏死黄脉病毒外壳蛋白。经光密度扫描估测,其表达量占大肠杆菌总蛋白的19.5％The temperature-inducible expression vector containing the beet necrotic yellow vein virus(BNYVV)isolate NM coat protein gene was constructed and transferred into E. coli DH5 α. High-level expression of the specific protein was achieved by temperature induction. The results of SDSPAGE and Western blotting show that the expression product which accumulates 19.5％ of the total cellular proteins estimated by Shimadzu CS-910 scanning is 21kd BNYVV coat protein.
