载脂蛋白B免疫遗传表型与高脂血症的关系

摘　　要：本文报告用抗人载脂蛋白B(apo B)单克隆抗体(B_5),检测不同个体等量低密度脂蛋白(LDL)的免疫反应性,发现其apo B均表现为高、中或低三种结合表型中的一种。遗传学调查结果证明,三种免疫反应表型变化系来源于apo B一对共显性等位基因(B_5^+/B_5^-)的遗传,B_5^-基因频率为0.593,B_5^-为0.407。血脂测定结果表明,高胆固醇血症组的B_5^+/B_5^-纯合子出现率明显低干正常组;在正常组内,B_5^+/B_5^+纯合子的血脂水平明显低于B_5^-/B_5^-纯合子,提示B_5^+等位基因似与低血脂,特别是低胆固醇有关。A monoclonal antibody against human apolipoprotein B (apoB),B_5,was used to test the immunnoreactivities of apo B in unrelated individuals. We found that the apo B in each person exhibited one of the three binding patterns: high,intermediate,or low affinity. Genetic investigation showed that these binnding patterns might result from co-dominant transmission of a pair of apo B alleles (B_5^+/B_5^-). The frequency of gene B_5^+ was 0.593 and B_5^-was 0.407. We also found that the frequency of B_5^+/B_5^+ homozygotes in the hypercholesterolemic patient group was significantly lower than that in healthy subject group. The serum levels of total cholesterol, triglycerides, very low density lipoprotein (VLDL)—cholesterol, and VLDL—triglyeerides in B_5^+/B_5^+ homozygotes were significantly lower than those in B_5^-/B_5^-in the group of healthy subject. It is suggested that allele B_5^+ might be associated with lower serum concentrations of lipids, especially the concentration of cholesterol.
