^(125)I标记人血白蛋白的HPLC分析和家兔体内代谢的研究

摘  要:用不同工艺生产的白蛋白及其^(125)I标记物(Sephadex G 50分离前后)均进行了HPLC紫外监测,标记蛋白同时作了放射分析。取分离后的标记白蛋白进行家兔体内代谢试验。结果表明,成都生研所柱层析、低温乙醇工艺和加拿大柱层析工艺产品的纯度分别为98.11%、99.38%和97.06%;其生物半衰期依次为96.11±0.01、90.00±1.69和85.75±2.62(小时),它们之间无显著性差异(P>0.05)。The UV monitoring of albumin (prepared by various technology) and ^(125) I-labelled albumin (before and after seperation by Sephadex G50) was carried out by HPLC. ^(125) I-labelled albumin was analysed by radioassay. Then the in vitro metabolic process of separated ^(125) I-labelled albumin in rabbits was studied. The result showed that the purities of albumin pripared by different technologies: column chromatography (Chengdu), cold ethanol (Chengdu) and column chromatography (Canada), were 98.11%, 99.38% and 97.06%, respectively; and the biological half life of them were 96.11±0.01, 90.00±1.69 and 85.75±2.62 hours, respectively. No significant difference was found among them.
