摘  要:本文报道的17羟孕酮(17-Hydroxyprogesterone,17-OHP)酶免疫荧光法。采用混合酐醛法制备17-OHP与辣根过氧化酶(HRP)结合物。利用羊抗兔IgG抗血清吸附的聚甲醛固相珠分离游离与结合的17-OHP-HRP。双氧水和对羟苯丙酸作为酶反应基质。通过荧光法测定酶的活性。该法的灵敏度为0.25~0.5pg/20μ1,组内、组间变异系数分别为5.00~7.95%,1.91~4.81%。在建立本法的同时,测定58例15~24孕周正常孕妇羊水,平均值45.71±26.56(5.22~120.42)pg/20μl,并且对一位高危先天性肾上腺皮质增生症(Congenital Adrenal Hyperplasia,CAH)产妇进行了孕中期羊水17-OHP测定,浓度为32pg/20μl,足月分娩一男婴,体检正常,出生72小时血清17-OHP正常,诊断为正常新生儿,与产前诊断符合。A method for enzyme immunoassy of 17-hydroxyprogesterone (17-OHP) was described. Horse-radish peroxidase (HRP) was used as the marker enzyme. The separation of the bound and free fractions was achieved by a second antibody coated with polyacetyl beads. 3-(p-hydroxyphenyl)propionic acid and H_2O_2 were used as the substrate and enzyme activity was measured fluorophotometrically. The assay sensitivity was 0.25—0.5 pg/tube. Within-assay C.V. was 5.00—7.95%. Interassay C. V. was 1.91—4.81%. The amniotic fluid value of 17-OHP investigated with this method in 58 samples of 15—24 week gestation amniotic fluid was 45.71±26.56 (5.22-120.41) pg/20μl. The concentration of 17-OHP measured in a second-trimester amniotic fluid sample obtained from a mother who had a high risk infant with congenital adrenal hyperplasia due to 21-hydroxylase deficiency was normal. After 40 weeks of gestation a normal male child was delivred.
