摘  要:本文建立了线性梯度洗脱,两波长切换检测和两内标法测定染毒大鼠尿中2苯甲酰肼叉-1.3二噻茂烷(BADYH)及其代谢产物的反相高效液相色谱方法.研究了大鼠ig不同剂量BADYH后,经尿排泄的DADYH及其代谢产物累积量-时间过程.结果表明,BADYH及其代谢产物累积排泄量占染毒总量的39.7%。其中主要代谢产物丙酮缩肝叉1.3二噻茂烷,占23.6%;肼叉1.2二噻茂炼占14.0%A simple,reliable reverse phase-high performance liquid chromatography method with linear gradient elution in two wave lengths and two internal standards was discribed for the simultaneous determination of benzoic acid-1,3-dithiolan-2-ylidenehydrazide(BADYH,Ⅳ)and its three principal metabolites: 1,3-dithiolan-2-ylidenehydrazine(Ⅰ),acetic acid-1,3-dithiolan-2-ylidenehydrazide(Ⅱ)and pyruvic acid-1,3-dithiolan-2-ylidenehydrazone(Ⅲ)in rat urine.The mean recoveries were more than 91%,the coefficients of variation were less than2.46%.BADYH and its metabolites have been determined in rat urine after 3 doses of BADYH.The experimental results show that the main metabolites excreted in rat urine are(Ⅲ)accounting for 23.6%,and(Ⅰ)accounting for 14.0%.The toxic mechanism of BADYH is also discussed.
