摘  要:本文用反相高效液相色谱法测定病人血浆中环丙氟哌酸浓度。以甲醇-乙腈-0.01 mol/L磷酸盐(25: 20:55)为流动相,荧光338/455nm检测。线性范围0.15~5.0μg/ml,平均回收率98.75%~104.32%,日内日间差异<8.18%。以本法测定10例呼吸道感染患者环丙氟哌酸血药浓度并分析药代动力学,结果单剂量口服500 mg环内氟哌酸后,药时曲线呈一级吸收二空模型。T_(max)=1.186±0.472h,C_(max)=2.573±1.198μg/ml。AUC=13.602±6.630μg/ml^(-1)·h,V/F=153.110±66.571L。T_(1/2)β=5.67±1.85h。Ciprofloxacin in serum was assayed by re-verse phase highpressure liquid chromatogra-phy with fluoresence detection. The mobilephase consisted of methanol, acetonitrile, and0.01mol/L phosphoric buffer.(25: 20: 55, pH=2.8). The linear range was 0.15~5.0μg/mland the recovery rate 98.75%~104.32%. Thecoefficient of variation of HPLC determinationwas less than8.18%. The pharmacokinetics ofciprofloxacin after oral administration of sin-gle doses of 500mg was determined in 10 res-piratory infection patients. The pharma-cokinetics of ciprofloxacin was shown by a twocompartement model. The mean peak serum concentration, time to peak concentration, andarea under the curve were 2.573±1.198μg/ml, 1.186±0.472h, and 13.602±6.630μg.ml^(-1) .h^(-1), respectively. The elimination half lifeand the apparent volume of distribution were5.67±1.85h and 153.110±66. 571L, respec-tively. A therapeutic regimen is suggested foruse of ciprofloxacin.
