摘 要:以生物素化dUTP标记CI(?)、株基因组Hind Ⅲ酶切Y片段为探针,用核酸杂交技术检测抗HCMV-iRNA制剂中巨细胞病毒核酸的残留,同时用HCMV免疫小鼠观察病毒核酸在小鼠体内的存留时间。结果,HCMV DNA在小鼠体内的存留时间有明显的个体差异,而与HCMV DNA有同源序列的RNA在免疫后7天被清除。在4批来自羊肝、脾组织的抗HCMV-iRNA 制剂中,有3批查出HCMV核酸。HCMVnuchic acid residues in the anti-HCMV immune RNA (iRNA)samph extracted from the sheep immunized with HCMV with detected by using DNA-DNA and DNA-RNA hybridization techniquea with the Biotin-labelled specific fragment of HCMV DNA as a probe. With this probe, 50pg of homologous NDA may be detected. The dynamic of HCMV nucleic acid was also observed in the mice after HCMV injection. The result showed that the RNA with the same homologous sequence as HCMV DNA could not be detected after 7 days of immunization. However the HCMV DNA in the body was largely various in the different indivaduals. 3 out of 4 samples of anti-HCMV iRNA from the sheep were showed to be HCMV nucleic acid positive.
