聚-L-赖氨酸-琼脂糖亲和色谱介质的研制及应用

摘　　要：用溴化氰活化法制备了聚-L-赖氨酸-琼脂糖亲和色谱介质。其配基的偶联量平均为2.5mg/ml胶,对猪胃蛋白酶的结合率为50mg/ml 胶。用其纯化猪胃蛋白酶,活性回收率在72%以上,提纯12倍。产品在 SDS-聚丙烯酰胺凝胶电泳中呈一条主带。Poly-L-lysine-QT_4 was prepared by coupling poly-L-lysine to QT_4 by the cyanogen bromide method.The mean amount of coupled poly-L-ly- sine was 2.5 mg per ml swollen gel.The binding capacity for porcine pepsin was approximately 50 mg per ml swollen gel.The affinity column was used to purify porcine pepsin.Pepsin was purified 12 times,with average activity re- covery over 72%,and was shown to be homogeneous by SDS-PAGE.
