氨苄青霉素钠锭剂、栓剂的人体药动学参数和生物利用度

摘  要:本文报告15例健康志愿受试者,其中甲组成人8例,乙组儿童7例,分别以医工院试制的氨苄青霉素钠锭剂口服,栓剂直肠给药,一次剂量均为250mg,1wk 后分别以苏州第三制药厂产品氨苄青霉素胶囊250mg 口服作自身对照。用药后于不同时间指端采血,用抑菌法测定血药浓度。实验结果表明锭剂的峰浓为胶囊剂的1.8倍,相对生物利用度为1.7。栓剂的达峰时间(t_(max)=0.25h)较胶囊剂(t_(max)=2.4h)快,峰值为胶囊剂的2.5倍。Fifteen healthy volunteers were divided into two groups.In group A,a 250mg oral dose of ampicillin sodium as trocheswas administered to 8 volunteers.In group B,a 250mg dose of ampicillin sodium as suppositories was administered by rectum to7 children.After 7 days,a250mg oral dose of ampieillin as capsules was administered to volunteers in each group as self-control.The serum concentrations of the drug were determined by microbiology assay.The pharmacokinetic parameters andbioavailability of the drug in troches and suppositories were compared with capsules respectively.The mean peak serumconcentration of the troches was 1.8μg/ml as high as the capsules and the relative bioavailability was 1.7.The time to peakserum concentrations of suppositories(t_(max)=0.25h)was quicker than capsules(t_(max)=2.4h).The mean peak serumconcentration of suppositories was 2.5 times higher than that of capsules.
