摘 要:新生大鼠胰碎片在MEM、DMEM、M-199和RPMI-1640中培养,比较培养不同天数冷冻保存,不同条件处理及液氮贮存不同时间的实验结果。表明新生大鼠胰碎片在RPMI-1640中培养胰岛素分泌量最高,随着培养时间的延长,胰岛素分泌量逐渐升高。培养6d进行冷冻保存为宜。新生大鼠胰碎片在液氮中贮存12个月分泌功能无明显变化。With pancreatic fragments from neonatal rats the effects of culture and freezing on the activities of islets were studied. The main results are as follows: The amount of insulin released during culture was higher in RPMI-1640 medium than that in DMEM, MEM or M-199 medium, increased after 2 days of culture, decreased on 4th day of culture and then gradually increased after 6 days. The amount of insulin released during culture after freezing-thawing correlated with the culture duration before freezing, increasing with increasing the culture time within a period of two weeks. The insulin release increased gradually during culture in all the three groups, i.e. culture group, freezing group and freezing after culture group. The ability of islets to secrete insulin was not changed after storage in liquid nitrogen for one week to one year.
