蛇床子总香豆精与挥发油和水提物对肾阳虚大鼠血浆前列腺素和环核苷酸的影响

摘　　要：为进一步研究蛇床子的补肾壮阳作用并筛选其有效成分,用醋酸氢化可的松制造大鼠肾阳虚模型,以蛇床子总香豆精、挥发油和水提物分别给药,测定大鼠血浆前列腺素(PGE_2和PGF_(2α))和环核苷酸(cAMP和cGMP)的浓度。结果阳虚组与正常组相比,PGE_2和PGF_(2α)显著降低(P<0.01),cAMP降低(P<0.01),cGMP相差不显著(P>0.05),但cAMP/cGMP值显著降低(P<0.01)。用药组总香豆精组和水提物组与阳虚组比较,PGE_2、PGF_(2α)及cAMP/cGMP值均显著升高(P<0.01)。用药组挥发油组与阳虚组比较,PGF_(2α)升高(P<0.01),但PGE_2和cAMP/cGMP值变化不明显(P>0.05)。通过实验认为影响体内环核苷酸和前列腺素代谢是蛇床子补肾壮阳作用的机理或途径之一,并认为香豆精成分是蛇床子补肾壮阳的作用的有效成分。Effects of total coumarins, essential oil and water extracts of Cnidium monnieri on plasmaprostaglandin (PGE_2 and PGF_2α) and cyclic nucleotide levels in rats of Kidney-Yang insufficiencywere studied. 55 rats were divided randomly into 5 groups, Group I was adminimstered orally withsaline (normal group), group Ⅱ was injected with intraperitonally hydrocortison acetate to induceKidney-Yang insufficiency (control group), group Ⅲ, group Ⅳ and group V (experimental groups)were injected with hydrocortison acetate, the same as group Ⅱ, and administered orally with thetotal coumarins; essential oil and water extracts of Fnictus Cnidii respectively, The levels of plasmaPGE_2, PG_2α and plasma cAMP, cGMP were measured. In group Ⅱ, in comparing with those of groupI, the levels of plasma PGE_2 and PGF_2α decreased significantly(P<0.01), and the vatue of cAMP/cGMP also lowered obviously(P<0.01) due to the significant reduction of cAMP and insignificantchange of cGMP. In group Ⅲ and group V, the above-mentioned indices changed significantly (P<0.01 or 0.05) compared with those of group Ⅱ, and after treatment it normalized basically incompar-ing with those of group I. In group Ⅳ, those indices didn't change regularily and apparently as groupⅢ and group V did, compared with group Ⅱ, and could not normaiize satisfactorily. It is suggestedthat the coumarins in the fruit of Cnidium monnieri are probably the effective ingredients toinvigorate Kidney and strenghten Yang, while the efficacy of essential oil remained unconfirmed.
