摘 要:本文报道抗纤溶剂戊烷脒从乙烯-醋酸乙烯酯(EVA)骨架体系中的释放行为。戊烷脒对EVA的渗透系数(D)及稳态流量(J_(lim))的测定结果表明释放机理是溶解控制释放。用EVA-琼脂骨架体系进行了溶胀实验,溶胀速率和溶胀程度随EVA的弹性模量而变化,较小的弹性模量得到较高的溶胀度从而导致较高的释放速度。弹性模量是影响EVA骨架体系药物释放速度的重要因素。Pentamidine isethionate (PI) was selected as an antifibrinolytic agent with a copolymer of ethylene -vinyl acetate (EVA) as its carrier. The steady state flux (J_(lim)) and diffusion coefficient (D) of PI through EVA membranes were determined using a permeation cell. The amount of PI diffusing through the membrane was smaller as compared with that from porous matrix. The release is dissolution controlled. A swelling test using EVA-agarose matrices were conducted to investigate the relationship of mechanical properties with the swelling. The swelling rate and degree are inversely proportional to the elastic modulus of EVA copolymers. The lower elastic modulus with the higher porosity of the matrix was obtained thus enhancing the drug release rate. The elastic modulus is a sensitive and basic factor in affecting the release kinetics.
